I've found that defense pierce is probably the most important thing here. I finished this using Medea Lily, Emiya, support Jalter, Georgios, Mozart, and Nightingale, with MLB white rose on Emiya and Jalter.
If you're going the Cu Alter route, the only thing I suggest is to try and kill everyone else before Medea and Ozy. Sure Hit ruins Cu Alter's entire gimmick so try to leave Ozy standing until the last minute. Yes, the guts Nero casts on death is annoying but way easier to deal with than what the other two throw at you.
